We present on novel drug designs rooted in physico-chemical principles with translational potential. We focus on two paradigmatic cases of target proteins, one with reported structure and the other where the structure is unknown. In the first case, a major cancer drug is re-engineered to synergize with immunotherapies and in the second case, a new lead to treat heart failure is discovered.
