Metoclopramide( MTC),a DA2 dopaminergic blocker reduces blood pressure in both normotensive and hypertensive subjects during cold presser test(M.Blanco et al Int J Clin.Pharmac Therap 34:390-392, 1996),treadmill exercise test(M.Blanco et al Amer. J.Therap.in press,1996).in the present study we are reporting a comparative evaluation between hypenensive (men and women)and defining MTC induced hypotensive action. Twenty hypertensive subjects(men and women) were studied.After I hour restingsubjects were submitted to the following experimental design: I)Two periods of placebo by using iv 5% glucose solution, before and after administration of MTC at the dose of 7.5uglKglmin.At the end of each period we measured blood pressure and heart rate prior, during and nfter handgrip test. Results:I )Hypertensive men:in placebo period I,HG increased BP from 158/102 to 174/119 and to 187/1!9 mmHg at I and 2 min,respectively .After MTC,pressor response to HG was 169/1I I and 176/102 mmHg,at I and 2 mirt,respectively. 2)Hypertensive women.in placebo period I,BP increased fmm 146/95 to 177/103 mmHg and to 192/101 mmHg,at land 2 min,respectively .After MTC infusion, blood pressure was reduced from 142/83 mmHg and during HG,the increment of I and 2 min,respectively. 162/97 mmHg to 168/91 mmHg at Conclusions: I)The hypertensive women exhibits greater reactivity to HG ,and 2) Hypotensive effect of MTC is more evident in women.

Nineteen hypertensive women were studied.After 1 hour resting,subjects were submitted to the following experimental design:l)Two periods of placebo by using iv 5% glucose solution. before and after administration of MTC at the dose of 7.5uglKg/min. At the end of each period we measured blood pressureand heart rate prior. during and after handgrip test. Results:l) Premenopausal women(n=9).During placebo period l,there was not any increase of blood pressure.During MTC period, the increase SBP was 20 and 18 mmHg at 1 y 2 min changes without in DBP;2)Postmenopausal women(n=l O).During placeb period I,handgrip increased systolic blood pressure by 3 I and 46 mmHg at I and 2 min,respectively .DBP increased 8 and 6 mmHg at I and 2 min,respectively .During MTC,pressor response to handgrip was reduced by 50?AThe increase of HR observed during p!accbo period I was attenuated during MTC and placebo period 11.The hypotensive effect induced by MTC was only noticed in premenopausal women. Conclusions:handgrip induces an exaggerated response and significantly geater in postmenopausal women and this increase is attenuated by iv MTC.
